Tubulin

Similar to Benzimidazole drugs that bind tubulin and are effective against parasitic worms and cancer.

Microtubule enlargement via calmodulin antagonism, Since calmodulin and calcium inhibit microtubule formation. But at higher levels of melatonin and in the absence of calmodulin and calcium, melatonin inhibited tubulin via non-specific binding to it [19].

Doesn't cause host Microtubule inhibition [20].

Deficiency

Conversion of Serotonin to Melatonin

Enzymes for conversion, SNAT (serotonin-N-acetyltransferase) which converts the serotonin to N-acetylserotonin, and HIOMT (hydroxyindole-O-methyltrasferase) which transfers a methyl group from S-adenosylmethionine to the 5-hydroxyl of the N-acetylserotonin [22]. So if you tend towards mania and ADHD you may have trouble converting serotonin to melatonin.

Retinoids deficiency can cause a deficiency in melatonin [23]

Vitamin d and melatonin are counter to one another. Increases in Vitamin D cause decreases in melatonin [24].

Linked to ADHD [25].

Zinc deficiency may be linked to melatonin deficiency and ADHD as well as issues with dopamine [26].

Dysregulated in hemochromatosis [27] or iron overload.

Deficiency of melatonin interacts with magnesium since magnesium is benzodiazepine GABA Agonist [28].

Risks

Melatonin is related to serotonin. Taking ssri's often causes an overgrowth of campylobacter which is killed with ginger. Campylobacter can cause insomnia so if melatonin stops working this may be why, that the melatonin is causing a campylobacter overgrowth. Take ginger.
